Molecular cloning generally uses DNA sequences from two different organisms: the species that is the source of the DNA to be cloned, and the species that will serve as the living host for replication of the recombinant DNA. Molecular cloning methods are central to many contemporary areas of modern biology and medicine. In a conventional molecular cloning experiment, the DNA to be cloned is obtained from an organism of interest, then treated with enzymes in the test tube to generate smaller DNA fragments. Subsequently, these fragments are then combined with vector DNA to generate recombinant DNA molecules. The recombinant DNA is then introduced into a host organism (typically an easy-to-grow, benign, laboratory strain of E. coli bacteria). This will generate a population of organisms in which recombinant DNA molecules are replicated along with the host DNA. HPLC chromatography lecture - This lecture explains about the HPLC chromatography technique in a nutshell by Suman Bhattacharjee. HPLC is performed to separate organic and biological compounds using solid stationary phase. High Performance Liquid Chromatography (HPLC) is a form of column chromatography that pumps a sample mixture or analyte in a solvent which is known as the mobile phase at high pressure through a column with chromatographic packing material known as stationary phase. The sample is carried by a moving carrier gas stream of helium or nitrogen. HPLC has the ability to separate, and identify compounds that are present in any sample that can be dissolved in a liquid in trace concentrations as low as parts per trillion. Because of this versatility, HPLC is used in a variety of industrial and scientific applications, such as pharmaceutical, environmental, forensics, and chemicals. Sample retention time will vary depending on the interaction between the stationary phase, the molecules being analyzed, and the solvent, or solvents used. As the sample passes through the column it interacts between the two phases at different rate, primarily due to different polarities in the analytes. Analytes that have the least amount of interaction with the stationary phase or the most amount of interaction with the mobile phase will exit the column faster. The main advantage of an uHPLC is speed. These systems are faster, more sensitive, and rely on smaller volumes of organic solvents than standard HPLC, resulting in the ability to run more samples in less time. It was widely used to diagnose infections, particularly with microbes that are not easily detected by culture methods, and in rheumatic diseases. However, in clinical diagnostics labs it has been largely superseded by other serological methods such as ELISA and by DNA-based methods of pathogen detection, particularly PCR. The complement system is a system of serum proteins that react with antigen-antibody complexes. If this reaction occurs on a cell surface, it will result in the formation of trans-membrane pores and therefore destruction of the cell. The basic steps of a complement fixation test are as follows:[1] Serum is isolated from the patient. Patients naturally have different levels of complement proteins in their serum. To negate any effects this might have on the test, the complement proteins in the patient's serum must be destroyed and replaced by a known amount of standardized complement proteins. The serum is heated in such a way that all of the complement proteins—but none of the antibodies—within it are destroyed. (This is possible because complement proteins are much more susceptible to destruction by heat than antibodies.) A known amount of standard complement proteins are added to the serum. (These proteins are frequently obtained from guinea pig serum.) The antigen of interest is added to the serum. Sheep red blood cells (sRBCs) [2] which have been pre-bound to anti-sRBC antibodies are added to the serum. The test is considered negative if the solution turns pink at this point and positive otherwise. If the patient's serum contains antibodies against the antigen of interest, they will bind to the antigen in step 3 to form antigen-antibody complexes. The complement proteins will react with these complexes and be depleted. Thus when the sRBC-antibody complexes are added in step 4, there will be no complement left in the serum. However, if no antibodies against the antigen of interest are present, the complement will not be depleted and it will react with the sRBC-antibody complexes added in step 4, lysing the sRBCs and spilling their contents into the solution, thereby turning the solution pink.
